Different extracts of Barringtonia asiatica (aqueous, methanol, petroleum, ether, hexane, acetone, methyl ethyl ketone and chloroform) were screened for anti-tumor activity (cytotoxic screening) utilizing topoisomomerases activity of mutant Saccharomyces cerevisiae. Zones of inhibition for all extracts were measured and recorded and only methyl ethyl ketone (MEK) and chloroform extracts of Barringtonia asiatica showed positive results against RS321 strain or topoisomerase, 1 zone of inhibitionwith an estimated 1C 12 values of 0.6458 mg/ml. 0.695 ml/ml and 0.75 mg/ml respectively. Rad 52 values for MEK and chloroform were both 0.85 mg/ml. A threefold difference between RS321 and Rad52 result is needed for a natural product to be regarded as having strong antitumor activity. Barringtonia MEK and chloroform extract showed positive results against Rad 321 for topoisomerase 1 activity due to an almost twofold difference between 1C12 values in Rad 52 and Rad 321.
